HepatoPac®Kits

Human HepatoPac® kits are an ideal way to conduct all your DMPK, Predictive Toxicology & Mechanistic Toxicology applications. Begin dosing as early as 2 days after receiving the kit! Use our Human HepatoPac® Triple Donor plates for the ultimate in convenience. HepatoPac® co-cultures derived from 3 independent and highly characterized donors are provided on a single plate (8-wells from each donor).

Human

HepatoMune™ Kits

HepatoMune™ is a highly functional in vitro liver platform that models inflammation-mediated hepatotoxicity. HepatoMune contains the proprietary patterning of hepatocyte “islands” in a “sea” of stromal with the addition of Kupffer cells to mimic the physiological microenvironment of the liver, enabling innate response to cytokine-mediated drug-induced liver injury. Its proven longevity of at least 10 days provides the experimental time needed to analyze the inflammatory response and its mechanisms of action.”

Human HepatoMune™ in vitro tri-cultures remain viable and highly functional for 10 days. These tri-cultures secrete albumin, synthesize urea, display functional bile canaliculi, and metabolize compounds using active Phase I and Phase II drug metabolism enzymes. The presence of functional Kupffer cells in HepatoMune™ tri-cultures is confirmed via pHrodo-S. aureus phagocytosis and CD68 staining. Primary hepatocyte function remains unaffected by the presence of activated Kupffer cells.

HepatoPac® B-CLEAR® Transporter Kits

B-CLEAR® technology ensures that the functioning hepatic transporters provide the most physiologic intracellular concentrations.

The HepatoPac® B-CLEAR® Transporter Kit combines the proven B-CLEAR® technology and contains Transporter Certified™ human hepatocytes, providing a standard for hepatocyte function and ensuring reproducibility from experiment to experiment. Transporter Certified™ hepatocytes have characterized uptake and efflux drug transport that result in physiologic intracellular concentrations, the driver for all intracellular processes. B-CLEAR® technology refers to a patented methodology that — when applied to properly functioning hepatic cells in culture — opens the bile pockets (analogous to bile canaliculi in vivo), allowing material that has been transported from inside the cell to be measured. Measuring biliary efflux provides the ability to evaluate biliary clearance, transporter interactions, and intracellular concentration. The B-CLEAR® technology is patented in over 45 countries worldwide and has been optimized over 10 years of work, ensuring that the primary hepatocytes are properly polarized with functioning uptake and efflux transporters. No other technology can deliver this level of transporter activity measurement, and therefore provide the most physiologic intracellular concentrations possible.
